ABSTRAK

EFEK IRADIASI SINAR GAMMA TERHADAP DAYA TAHAN BAKTERI Pseudomonas aerugillosa DALAM BERBAGAI KONDISI.

Telah dilakukan penelitian efek radiasi sinar gamma 60Co terhadap bakteri P. aeruginosa dengan tujuan untuk mengetahui nilai D10 bakteri yang diiradiasi dalam berbagai kondisi. Sampel bakteri dari suspensi akuades, talek dan tepung kacang tanah dalam kondisi kering, basah, 02 dan N2, diiradiasi pada sinar gamma dengan dosis 0 - 2,5 kGy dan laju dosis 5 serta 10 kGy/jam. Setelah diiradiasi sample bakteri P. aeruginosa dibiakkan pada media Tryptotle Soya Agar, selanjutnya diinkubasi pada 32 ± 2 °C selama 3 hari. Koloni yang tumbuh dihitung, datanya dipakai untuk membuat kurva dan menentukan nilai D10. Hasilnya menunjukkan bahwa tidak ada perbedaan berarti m1tara nilai D10 bakteri yang diiradiasi dalam suspensi akuades, talek dan tepung kacang tanah. Nilai D10 bakteri yang diiradiasi pada laju dosis 10 kGy/jam cenderung lebih tinggi daripada yang diiradiasi 5 kGy/jam. Nilai D10 mikroba yang diiradiasi dalam kondisi N2 lebih tinggi dari pada dalam kondisi O2.

EFFECTS OF GAMMA RAYS IRRADIATION ON RESISTANCE OF Pseudomonas

aerugillosa BACTERIA IN VARIOUS CONDITION.

The investigation of ganuna rays 60Co irradiation effect on resistance of bacteria P. aeruginosa has been done. The objective of the research was to know the D10 value of bacteria P. aeruginusa. By using of distilled water, talc and peanut powder as carrier in dry, wet, 02 and N2 condition the bacteria of P. aeruginosa were irradiated on gamma rays of 60 Co with dose of 0 to 2.5 kGy, and with dose rate of 5 and 10 kGy/h. After irradiation the bacteria of P. aeruginosa were cultured in media of the Tryptol/e Soya Agar and incubated at temperature of 32 ± 2 °C for 3 days. The survival colonies were calculated, and the data were used to make the curve and to determine the  D10 value. The results of the experiment showed that D10 value of irradiated bacteria of P. aeruginosa in the distilled water, talc and peanut powder as carrier were not high significant. Nevertheless the D10 value of the irradiated at dose rate 10 kGy/h show more higher tendency than at dose rate 5 kGy/h. The D10 value of irradiated bacteria in the N2 condition was higher, if compared with in the 02 condition.
